Sensitive detection of rare somatic mutations

Boreal’s OnTarget™ allele enrichment system enables detection of rare somatic mutations that are present at frequencies down to 0.01% relative to wild-type and background DNA, enabling identification of tumor DNA in plasma; frozen, FFPE or fresh tissues; and cell lines. The key features of the product are:

OnTarget™ allele enrichment will allow clinicians and oncology researchers to develop highly sensitive assays that will help guide clinical decisions during targeted therapy regimens and to help determine earlier when treatment should be altered due to emergence of drug resistance.

OnTarget™ enrichment for the BRAF V600E mutation demonstrates >1,000,000x reduction of the wild-type while maintaining a high yield of the mutant.

Dramatic improvement in sensitivity gained by adding OnTarget™ to a qPCR based mutation detection workflow. In both panels, the assay is challenged with a mixture of BRAF V600E mutant cell line diluted to different amounts into BRAF wild type cell line. At 7% mutation abundance, the qPCR assay alone is barely capable of distinguishing the mutant above the negative control, while at 0.03%, qPCR cannot distinguish the mutant containing sample from the negative control.

By adding OnTarget™ in the workflow, even 0.03% mutant abundance is easy to distinguish above the negative control, demonstrating a 200-fold increase in sensitivity.
